NuCLEANase® for Viscosity Reduction of Fermentation Broths

In fermentation processes, NuCLEANase® can be used to reduce the nucleic acid induced viscosity in fermentation broths, enabling a significantly more efficient product recovery.

The strong tendency of DNA molecules to increase viscosity arises from their molecular characteristics. The great length of DNA strands causes them to entangle with one another, leading to increased viscosity. In addition, the negatively charged phosphate backbone is surrounded by a layer of less dynamic water molecules, further contributing to the viscosity.

Challenge
High viscosity of fermentation broths greatly impacts the efficiency the recovery process. The Separation of solids from the broth by centrifugation or filtration becomes less effective due to the increased resistance of the fluid, requiring more energy for pumping. Also, a faster fouling of membranes is often seen with higher viscosities.

Solution
NuCLEANase® is a simple and efficient solution for the reduction of nucleic acid induced viscosity. The addition of 10 kU/ml of NuCLEANase® to the culture broth of a large-scale microbial Bacillus sp. Fermentation reduced the viscosity of the broth by 88% within one hour after addition of the enzyme. As a result of the lower viscosity the transmembrane pressure during the following micro-filtration step was lower over a longer time and easier to control. This resulted in a 22 % higher overall throughput and a reduction of the filtration time by 7 hours (25 %).
